In Russia the largest and various library of human antibodies in the world is created and announced in November, 2015. It includes blood samples more than one thousand donors (MeganlibTM). Researchers of biotechnology corporation BIOCAD bring together library since 2012. Genes of antibodies which are stored here serve for creation of drugs of new generation, from oncological, autoimmune and other diseases. First of all, it is medicines on the basis of bispetsifichesky monoclonal antibodies on which scientists of the world work now.

The library represents storage where there are billions of genes of different antibodies, each of which is individual and capable to contact a certain molecule target. Libraries of antibodies happen three types: the naive, consisting of human donor samples, synthetic, i.e. synthesized de novo from oligonukleotid, and immune, received from blood of animals. MEGAN Libbiblioteka of monoclonal antibodies treats the first type and it is created in a special format for selection of antibodies by method of the fagovy display. Probability of effective and safe treatment by the medicines developed on the basis of the antibodies received from donor human libraries, above than in case of the synthetic or humanized antibodies.

As a rule, libraries of human antibodies total about ten donors, in rare instances – to several hundred, for example, in such companies as Pfizer and Medimmune. MorphoSys designed several libraries with a big variety of material, but samples in them synthetic. BIOCAD has the biggest and various library of the antibodies in the world generated by the immune system of the person. Now the company works with library of 1,100 donors, and already in the next half a year is going to complete collecting of samples, having increased their number up to 2,000.

Testing of antibodies is held in special robot stations. On average, about 100 thousand samples are during the day processed. A meeting and the maintenance of such library of antibodies (MeganLiB) – the procedure technically difficult and labor-consuming. The staff of three laboratories is engaged in process of support and replenishment of library of human antibodies of BIOCAD. Information on each sample without fail is registered in electronic journals of accounting and also is displayed in protocols which are filled out by responsibles of laboratories after each stage of work with library. Materials of library are stored at low temperatures in refrigerators in the following formats: lizirovanny fraction of mononuclear cells of peripheral blood, RNA and KDNK. RNA and KDNK are located on storage in the form of two alikvot.

A variety of material allows to find the necessary antibodies for molecules targets on a surface of a cancer cell in library. From huge number of antibodies researchers select only several tens of the molecules capable to neutralize the protein contributing to the development of cancer or to activate immune cages of an organism on destruction of a tumor. Thus the innovation medicines are developed.

The library looks as a small two-millilitre test tube with liquid, it, nevertheless, contains all power of the immune system thousands of people, in the form of one hundred billions of various genes coding so-called, Fab fragments of antibodies.

The main need of creation of so various donor library - the big repertoire of different structural options. It considerably increases the probability of creation of medicine with the set therapeutic properties. Development of medicines on the basis of monoclonal antibodies – one of the most relevant and difficult technologies of modern pharmaceutics. According to the statistics, every third person suffers from any malignant new growth. Monoclonal antibodies beat precisely cancer cells, making the minimum impact on healthy, unlike the same chemotherapeutic medicines.

The library of human antibodies MEGANLIB is used in all projects of BIOCAD company, first of all, when developing the innovation medicines within the MabNext program. Now the company keeps more than twenty projects. The value of this library in its universality, thanks to it scientists can find an antibody on any target which interests them. It can be also targets for treatment of infectious diseases. Besides the detected antibodies which are specifically contacting markers of different diseases can be used in diagnostics. Perhaps, in the future the genetic material of cages of the immune system collected in MEGANLIB library can be necessary in the field of basic researches for creation of the database of antibodies and genes.
